If I jailbreak an iPhone will I not be able to update my iPhone to latest iOS.
Also will it affect my phone in any way such as not being able to use app store or iTunes

If you can, update to the latest firmware which is iOS 5.0.1 and it will be untethered. And no it will not effect your phone in ANYWAY!

If you update you have to restore and jailbreak it again on that different iOS

Just stay on the current OS until the next one is jailbroken.

Everytime there is an update you will have to remove the jailbreak in order to update your iPhone.
