You will more than likely get this just by playing through the story as there are way more than 50 compys for you to defeat. A few levels actually have compy goons that will continuously spawn until you block the area they come out with, meaning you could theoretically kill all 50 in a single level. If you dont have this trophy unlocked by the end of the story then there are still multiple gold bricks in free roam that will require you to kill compys.
