Before you start going for this, you will want to have completed the single player and the co-op challenges. As you will already have a good amount of kills.
You will want to do this in Sand Box mode, but make sure you are the host, as only the host's kills will count.
The best way to do this is to get an SUV, and add a few things to it.
Firstly, you want to get the two Gambling mags. One located on the second floor of the BBQ shack in the Americano Casino. And the second located on the second floor of Grotto in the Palisades.
Once you have these two things, go to the Slot Ranch Casino. In the casino go to the large slot machine, and keep playing until you earn $2,500,00.
Once you have earned the money, visit the Pawn Shop on the Silver Strip and purchase the SUV key.
After that, you will want to find the Driving Magazine so your SUV will be able to take more damage and ultimately last longer. You will find this at the Paradise Platinum Screens on the Platinum Strip.
Find your way to the roof, you can do so by jumping onto the candy machine located by Cash Gordan's Casino. You want to climb up, follow the hallway that leads upward. And finally you will jump to the roof of Jugz Bar and Grill, over to Paradise Platinum Screens.
Once you have the magazine, you will want to go and get the SUV, from then on you will simply run over zombies up and down the strip until your SUV begins to break. You will know when it begins to break as you will start to see smoke.
At this point, go near the SUV spawn and wait until the one you are using breaks, and get a new one.
Continue to do this until you reach the kill count, take note that this may take some time.
